There is no way but by using triggers like set Heroes mana=mana -300 or such operation I think...:(

If it's the only spell the unit has, just modify the mana capacity of the unit (so if you want a spell to require twice the default amount of mana, divide the mana capacity of the unit by two instead).. there's no other way to do it.
You can edit the ability.slk file with excel or Microsoft Works and change the mana cost. But the problem is it will cause desync in multiplayer and drop people in the game, but if your planning is use in a single player game it would work if included on a custom MPQ.
