Yes and no, you can simulate multiple floors with some complex destructable stuff for 1 player, but if you put more than 1 player into the mix or units that move around the building then WCIII goes NO FUCK U.

Toadcop does it with his shooter map. I think his has something to do with a physics engine, and then creating an "object" that acts like a solid surface for the engine.

You can also simulate it with flying heights. Just make it so when the unit leaves the stairs or w/e, set its flying height to the height of the floor.
